Although DFAST can be used without user registration, you can also log-in to the web service with a user account for D-way, a data submission portal to DDBJ. While logged-in, the Job History page becomes available.

In the Job History page, you can browse your jobs executed while you are logged-in. Bulk data downloading for multiple job results and batch job deletion are also available. You can import a job to the history by specifying the job ID.

DFASTはユーザー登録をせずに使用することもできますが、DDBJの登録ポータルであるD-wayのユーザーアカウントを使ってログインすることも可能です。ログインするとジョブ履歴の管理画面が利用できるようになります。

ジョブ履歴管理画面では、ログイン時に実行したジョブの一覧が閲覧でき、ファイルの一括ダウンロードや、一括削除などを行うことができます。ログインしていない状態で実行したジョブについては履歴に表示されませんが、実行済みのジョブのIDを指定して履歴に取り込むこともできます。

D-wayアカウントはこちらのページで取得することが可能です。DDBJへのゲノム塩基配列登録時に必要なBioProjectやBioSampleの登録にもD-wayを用いますので、DDBJへのデータ登録を予定している方にはアカウントの登録をおすすめします。

1. Registration for BioProject / BioSample / Locut Tag Prefix

In prior to data submission to DDBJ, you have to register BioProject Database and BioSample Database to obtain accession numbers. Optionally, raw read sequences should be deposited in Sequence Read Archive (SRA) Registration can be done at D-way, a data submission portal of DDBJ.

Locus Tag Prefix is also required for submitting the annotated genome sequences. Locus Tag Prefix is used as a prefix for the gene locus identifier. You cannot use the same prefix as the ones used in other genomes already deposited in INSDC.

2. Run DFAST

Run DFAST and fill out the metadata form in the "DDBJ Submission" tab in the result page. BioProject/BioSample IDs and Locus Tag Prefix registered in the above step should be specified here.

After providing the metadata, press "Format Check" button to validate the metadata. Errors (ER1 or ER2) must be corrected if any. Warnings (WAR) might need correction before submitting the data. Please refer to this pagefor the details of the error message.

3. Submitting the data to DDBJ

Data submission files can be downloaded either by pressing the "Zip Download" button in the "DDBJ Submission" tab or from the job history page, which is available when you are logged-in to DFAST with a D-way user account.
In the job history page, select jobs that you want to submit and choose the file format "MSS" for data submission files, then press "DOWNLOAD" button.

The downloaded files can be submitted to DDBJ from MSS (MASS Submission System) form.

1. BioProject / BioSample / Locut Tag Prefix の取得

DDBJへのゲノム塩基配列データ登録に際して BioProject Database と BioSample Database への登録を行い、アクセッション番号を取得する必要があります。 また、必要に応じてシークエンスデータの raw data の SRA への登録も行ってください。 いずれもデータ登録ポータル D-way で登録を行うことができます。各ゲノムに対しBioSampleをそれぞれ登録する必要があります。１つの研究プロジェクトの中で複数のゲノム配列を決定した場合には１つのBioProjectに対して複数のBioSampleを紐づけることができます。

アノテーションされたゲノム配列の登録には、BioProject もしくは BioSample 登録時にLocus Tag Prefix を取得する必要があります。 Locus Tag Prefix は遺伝子座を示す識別子の prefix として用いられるものです。すでに INSDC に登録済みのゲノムで使われている prefix を重複して使用することはできません。

2. DFASTの実行とメタデータの入力

DFAST を実行し、結果画面の "DDBJ Submission" タブにおいて必要なメタデータ (登録者や生物情報) を入力します。1.で取得したBioProject・BioSampleやLocus Tag Prefixもここで指定します。

メタデータ入力後、"DDBJ Submission" タブ内にある "Format Check" ボタンを押して登録ファイルに問題がないかチェックを行い、エラー (ER1やER2など)があれば修正を行ってください。警告(WAR)については修正が必要になる場合があります。 エラーメッセージについてはこのページをご参照ください。

3. 登録ファイルのダウンロードとDDBJへの提出

登録用ファイルは"DDBJ Submission"内の "Zip Download" ボタンまたは、ログイン時に表示されるジョブ履歴管理画面からダウンロードできます。 ジョブ履歴管理画面からダウンロードを行う場合には、対象となるジョブをチェックして選択し、ファイル形式として "MSS" を選んだ後、"DOWNLOAD"ボタンを押してください。

MSS申し込みフォームからMSS (大量登録システム) の利用申請を行い、フォームの指示にファイルのアップロードを行ってください。 （新MSSフォームは近日リリース予定：当面はメール等でのファイル送信を行ってください）
